Semaglutide mimics GLP-1, a hormone the body releases after eating. It acts on GLP-1 receptors involved in appetite regulation and slows how quickly the stomach empties, which may help some patients feel full sooner and reduce food intake. Responses vary from person to person.

GLP-1-related treatment requires an individualized risk assessment and ongoing medical follow-up. This is not a complete list — your provider reviews all risks with you.

  • Nausea, vomiting, diarrhea, constipation, and abdominal discomfort are the most commonly reported side effects, especially when starting or adjusting a dose.
  • Boxed warning: in rodent studies these medications caused thyroid C-cell tumors. They should not be used by anyone with a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).
  • Serious risks can include pancreatitis, gallbladder problems, kidney injury related to dehydration, serious allergic reactions, and low blood sugar when combined with insulin or sulfonylureas.
  • Changes in mood or thoughts of self-harm should be reported to a provider right away.
  • These medications are not recommended during pregnancy or while trying to conceive, and may affect how some oral medications are absorbed.
  • Seek medical care immediately for severe abdominal pain, signs of an allergic reaction, or symptoms such as a lump or swelling in the neck, trouble swallowing, or persistent hoarseness.
  • Semaglutide: Patients with diabetes should have any history of diabetic retinopathy reviewed, as rapid improvement in blood sugar has been associated with retinopathy complications.
